4、免疫組化染色步驟：
石蠟切片脫蠟至水。
3%H2O2室溫孵育5~10 分鐘，以消除內源性過氧化物酶的活性。
蒸餾水沖洗，PBS 浸泡5 分鐘，(如需采用抗原修復，可在此步后進行)。
5~10％正常山羊血清（PBS 稀釋）封閉，室溫孵育10 分鐘。傾去血清，勿洗，滴加適當比例稀釋的一抗或一抗工作液，37℃孵育1~2 小時或4℃過夜。
PBS 沖洗，5 分鐘×3 次。
滴加適當比例稀釋的生物素標記二抗（1%BSA-PBS 稀釋），37℃孵育10~30 分鐘；或滴加第二代生物素標記二抗工作液，37℃或室溫孵育10~30 分鐘。
PBS 沖洗，5 分鐘×3 次。
滴加適當比例稀釋的辣根酶標記鏈霉卵白素（PBS 稀釋），37℃孵育10~30 分鐘；或第二代辣根酶標記鏈霉卵白素工作液，37℃或室溫孵育10~30 分鐘。
PBS 沖洗，5 分鐘×3 次。
顯色劑顯色（DAB 或AEC）。
自來水充分沖洗，復染，封片。
冰凍切片免疫組化染色步驟
冰凍切片4~8?m，室溫放置30 分鐘后，入4℃丙酮固定10 分鐘，PBS 洗，5 分鐘×3。用3％過氧化氫孵育5~10 分鐘，消除內源性過氧化物酶的活性。
PBS 洗，5 分鐘×2。
4, immunohistochemical staining steps:
Paraffin sections dewaxed to water.
3% H2O2 incubated at room temperature for 5 to 10 minutes to eliminate endogenous peroxidase activity.
Rinse with distilled water and soak in PBS for 5 minutes. (If antigen retrieval is required, follow this step.)
5 ~ 10% normal goat serum (diluted in PBS), incubated for 10 minutes at room temperature. Decant the serum, do not wash, dropping the appropriate proportion of diluted primary antibody or a working solution, incubated for 1 to 2 hours at 37 ℃ or overnight at 4 ℃.
Wash in PBS for 5 minutes × 3 times.
(1% BSA-PBS diluted), incubated at 37 ℃ for 10-30 minutes; or the second generation of biotin labeled secondary antibody working solution, incubated at 37 ℃ or room temperature for 10 to 30 minute.
Wash in PBS for 5 minutes × 3 times.
Add streptozotocin labeled with horseradish enzyme diluted in appropriate proportion (diluted in PBS) for 10-30 minutes at 37 ° C, or streptozotocin working solution labeled with horseradish peroxidase at 10 ° C minute.
Wash in PBS for 5 minutes × 3 times.
Color developer color (DAB or AEC).
Full flushing water, counterstainer, sealing piece.
Frozen section immunohistochemical staining procedure
Frozen sections 4 ~ 8? M, at room temperature for 30 minutes, into 4 ℃ acetone fixed 10 minutes, PBS wash, 5 minutes × 3. Incubate with 3% hydrogen peroxide for 5 to 10 minutes to eliminate endogenous peroxidase activity.
Wash with PBS for 5 minutes × 2.
